Вопросы: ДНК-технологии в животноводстве. Трансплантация, трансгенез и клонирование в животноводстве.

Биотехнология в животноводстве включает следующие направления:

1) создание генно-модифицированных (трансгенных животных) с целью улучшения их генетических характеристик как продуцентов молока и мяса, либо с фармацевтическими целями (получение терапевтических белков в коровах и козах);

2) применение молекулярно-генетического маркирования для отбора лучших особей в при племенном разведении;

3) клонирование животных посредством ядерного переноса из соматических клеток, что позволяет генетически реплицировать отобранных животных.

История трансгенеза у животных. Richard Palmiter developed the techniques responsible for transgenic mice, rats, rabbits, sheep, and pigs in the early 1980s. The first transgenic (genetically modified) animal was produced by injecting DNA into mouse embryos then implanting the embryos in female mice. Herman the Bull was in 1991 the first genetically modified or transgenic bovine in the world. The announcement of Herman's creation caused an ethical storm.

Цели трансгенеза у животных. Genetically modified animals currently being developed can be placed into six different broad classes based on the intended purpose of the genetic modification:

- to research human diseases (for example, to develop animal models for these diseases);

- to produce industrial or consumer products (fibres for multiple uses);

- to produce products intended for human theraputic use (pharmacutical products or tissue for implantation);

- to enrich or enhance the animals' interactions with humans (hypo-allergenic pets);

- to enhance production or food quality traits (faster growing fish, pigs that digest food more efficiently);

- to improve animal health (disease resistance).

Методы трансгенеза у животных:

(1) микроинъекции ДНК в зиготу;

оплодотворенные яйцеклетки извлекают из организма животного и помещают в культуру (в питательный раствор), поддерживают в микрокапельной культуре в CO2-инкубаторе (5% CO2) при 37С. Все остальные манипуляции с зиготами производят в специальной забуференной среде: в них с помощью микроманипулятора вводят клонированную ДНК любого размера. Для пересадки зигот, в которые произведена микроинъекция, используют «суррогатных» матерей – псевдобеременных самок, которых спаривали со стерильными самцами. Часть пересаженных зигот погибает. После рождения детенышей или получением их посредством кесарева сечения, проводится генетический анализ трансформантов: выделяется ДНК из маленьких фрагментов хрящевой ткани детенышей и проводится ее гибридизация с пробой. Трансгенные линии животных получают после скрещивания первичных трансгенных экземпляров.

(2) заражение предимплантированных эмбрионов рекомбинантными ретровирусами (трансфекция);

схема технологии: введение чужеродного гена в геном вируса (получение вирусного вектора) –> инфицируют культуру фибробластов –> помещают 8-клеточную морулу (стадия дробления зиготы) в такую культуру –> морула инфицируется –> ее вводят в матку псевдобеременной самки –> часть бластоцист погибает, а выжившие и развившиеся в детенышей ―> подвергаются генетическому анализу ―> выводятся трансгенные линии.

(3) применение эмбриональных стволовых (ES- и EK-)клеток.

плюрипотентные эмбриональные стволовые клетки выделяют из бластоцист, культивируют некоторое время in vitro ―> в них вводятся клонированные гены путем инфекции или трансфекции (через вирусный вектор) ―> вводят трансформированные клетки назад в бластоцисту ―> трансформированные стволовые клетки колонизируют эмбрион ―> успешные трансгенные детеныши отбираются через молекулярно-генетический анализ.

Практические примеры использования трансгенеза в животноводстве:

- получение быстро растущих трансгенных свиней;

- The enviropig is the trademark for a genetically modified line of Yorkshire pigs with the capability to digest plant phosphorus more efficiently than ordinary unmodified pigs that was developed at the University of Guelph. Enviropigs produce the enzyme phytase in the salivary glands that is secreted in the saliva; The enzyme was introduced into the pig chromosome by pronuclear microinjection. With this enzyme, Enviropig is able to digest cereal grain phosphorus, so there is then no need to supplement the pigs' diet with either phosphate minerals or commercially produced phytase, and less phosphorus is lost in the manure;

- выведение трансгенных овец, вырабатывающих в молочных железах некоторые факторы свертываемости крови человека; On 6 February 2009, the U.S. Food and Drug Administration approved the first human biological drug produced from such an animal, a goat. The drug, ATryn, is an anticoagulant which reduces the probability of blood clots during surgery or childbirth. It is extracted from the goat's milk;

- In 2011, Chinese scientists generated dairy cows genetically engineered with genes for human beings to produce milk that would be the same as human breast milk;

- In 2012, researchers from New Zealand also developed a genetically engineered cow that produced allergy-free milk;

- BioSteel is a trademark name for a high-strength based fiber material made of the recombinant spider silk-like protein extracted from the milk of transgenic goats, made by Nexia Biotechnologies. The company has successfully generated distinct lines of goats that produce in their milk recombinant versions of either the MaSpI or MaSpII dragline silk proteins, respectively.

- In 2006, a pig was engineered to produce omega-3 fatty acids through the expression of a roundworm gene;

- Генная инженерия в рыбоводстве пока очень мало развита. There is a genetically altered salmon with an increased growth rate being considered for FDA approval. Genetically modified fish have been developed with promoters driving an over-production of growth hormone for use in the aquaculture industry to increase the speed of development and potentially reduce fishing pressure on wild stocks. AquaBounty, a biotechnology company working on bringing a GM salmon to market, claims that their GM AquAdvantage salmon can mature in half the time it takes non-GM salmon and acheives twice the size. AquaBounty has applied for regulatory approval to market their GM salmon in the US. As of May 2012 the application was still pending.

 

Трансплантация у животных: технология пересадки эмбрионов от различных животных, с получением полноценного потомства, была известна с конца XIX в. Такого рода манипуляции с зиготами или эмбрионами относят в разряд клеточной инженерии, и, как более простую технологию, не всегда относят в разряд биотехнологии. Пересадка эмбрионов известна в практике животноводства на: коровах, овцах, козах, свиньях, кроликах. В лаборатории применяется на мышах, в целях фундаментальных исследований.

Для получения элитного стада коров за относительно короткий интервал времени: отбирают родоначальницу ―> вводят фолликулстимулиющий гормон ―> гормон вызывает выброс 10-15 яйцеклеток вместо 1 ―> яйцеклетки оплодотворяются ―> через 4-5 дней 32-клеточные, неприкрепившиеся эмбрионы вымывают ―> определяют их полноценность ―> вводят микрошприцем непородистым коровам, которым ввели простагландины для стимуляции воспроихводительного цикла ―> коровы-реципиенты вынашивают и рождают элитных телят. Темпы создания стада: от одной элитной коровы в среднем получают элитных 5 телят за 15 месяцев.

Вариации данной технологии: оплодотворение in vitro, а также создание фонда замороженных эмбрионов, которые можно продавать в другие хозяйства, где нет элитной коровы и необходимой техники, кроме инструментов для подсадки эмбрионов не-элитным коровам.

Клонирование сельскохозяйственных животных. Под термином клонирование у животных подразумевают клонирование организма (син. репродуктивное клонирование, бесполое размножение).Определение: р.к. – это получение многоклеточного организма, генетически идентичного другому организму, без оплодотворения.

Клонирование у животных концептуально отличается от клонирования у растений и грибов.

Репродуктивное клонирование у животных производят 2-мя способами:

(1) пересадка ядра из соматической клетки - "somatic cell nuclear transfer" (SCNT). Источником для создания клона у скота являются обычно клетки кожи ушной раковины. Из зрелой (дифференцированной) клетки животного-донора извлекают ядро и вводят в яйцеклетку, лишенную ядра. Выжившуу после такой операции яйцеклетку имплантируют суррогатной матери. Ограничения: такая процедура, тем не менее, не всегда создает генетически идентичную копию, так как соматическая клетка может иметь индивидуальные мутации ядерной ДНК, отличающие ее от клеток остальной части организма, или быть частью химеры – тканевой мутации. Кроме того, митохондриальный геном будущий сконструированный организм получает только из реципиентной яйцеклетки. Иногда несовместимость ядра и митохондрий приводит к гибели подобной генетической конструкции.

(2) искусственное дробление эмбриона на 6-8-клеточной стадии. Так получают монозиготических близнецов.

История клонирования животных.Первые шаги к репродуктивному клонированию у животных были сделаны еще в 1928 Otto Mangold произвел пересадку ядра из соматической клетки лягушки в эмбрион лягушки.

Овца была наиболее ранним объектом для разработки клонирования. В 1986 овца была получена из клеток раннего эмбриона, в 1995 – из дифференцированных эмбриональных клеток.

В период 1996-2009 из соматической клетки были впервые клонированы овца (1996), корова (1997), свинья (2000), коза (2001), лошадь (2003), мул (2003), кролик (2003), верблюд (2009).

Статистические данные по эксперименту с пересадкой ядра из клетки вымени в яйцеклетку у овцы (1996): Cloning Dolly the sheep had a low success rate per fertilized egg; she was born after 277 eggs were used to create 29 embryos, which only produced 3 lambs at birth, only 1 of which lived.

Possible Abnormalities due to Cloning.Researchers have found several abnormalities in cloned organisms, particularly in mice. The cloned organism may be born normal and resemble its non-cloned counterpart, but majority of the time will express changes in its genome later on in life. The theory behind this is that the biological blueprint of the genes is the same in cloned animals as it is in normal ones, but they are read and expressed incorrectly. Results of these abnormally expressed genes in the cloned mice were premature death, pneumonia, liver failure and obesity.

Перспективы развития генетической инженерии. Некоторыепредсказания футурологов:

- 2011-2016 - 20%+ efficient genetically engineered algae ponds to generate hydrogen;

- scientists are taking an artificial chromosome containing the gene to correct a serious disease and are simply putting it in a stem cell and transplanting into the body;

- First (now-2015) we will see more and more smart drugs, i.e. drugs increasingly designed based on understanding on how genes and proteins work and not just random hit-and-miss experiments with organic molecule;

- Then (2015-2025) we are going to see first health-related genetic modifications. As e.g. Naam describes in his book, there are several ways to modify ourselves - inject the protein (drug - see above), add the DNA into the cell (noninsertional vectors) or into the nucleus (insertional). By that time we will strongly feel the fallout from the Human Genome project, understanding a signficant part of the complex genetic chemistry;

- At the same time we will have genetic treatments in competitive sport (Olympics) to enhance the strength, endurance, etc. Some say that first such treatments are already being used. Each next Olympics will have more and more and by 2016 a very large portion of athletes will be using some form of genetic enhancement;

- By 2030 most people will have the ability to design new organisms from scratch and change their own bodies at will.

Генетическое оружие. (Синонимы: биогенетическое оружие, этническое биооружие). Этническое или биогенетическое оружие – нацеленное во вред только или в первую очередь каким-либо индивидам, этносам или генотипам.

History. In 1997, U.S. Secretary of Defense William Cohen referred to the concept as a possible risk.

The Next Battle in Washington. Social Security Mobilizes Troops. By James Ridgeway Tuesday, Jan 26 1999. Adding to press accounts of Israeli research into an ethnic bomb came a report, "Biotechnology, weapons and humanity," last week from the esteemed British Medical Association, warning that "weapons could theoretically be developed which affect particular versions of genesclustered in specific ethnic or family groups." It adds: "As genetic manipulation becomes a standard laboratory technique, there is a risk that this new information will also become widely available and procedures to monitor against the misuse of this new knowledge are urgently needed."

The possibility of a "genetic bomb" is presented in Vincent Sarich's and Frank Miele's book, Race: The Reality of Human Differences, published in 2004. The authors believe that information from the Human Genome Project will be used in just such a manner. In 2005 the official view of the International Committee of the Red Cross was "The potential to target a particular ethnic group with a biological agent is probably not far off. These scenarios are not the product of the ICRC's imagination but have either occurred or been identified by countless independent and governmental experts." Israeli "ethno-bomb" controversy: In November 1998, The Sunday Times reported that Israel was attempting to build an "ethno-bomb" containing a biological agent that could specifically target genetic traits present amongst Arab populations. In 2007, Russian newspaper Kommersant reported that the Russian government banned all exports of human biosamples. The report claims that the reason for the ban was a secret FSB report about on-going development of "genetic bioweapons" targeting Russian population by Western institutions. The report mentions the Harvard School of Public Health, American International Health Alliance, United States Department of Justice Environment and Natural Resources Division, Karolinska Institutet and United States Agency for International Development.
